Cyclodextrin Platforms for Sensitive Detection of an Environmental Exposure Biomarker
Introduction
Assessing exposure to environmental chemicals requires accurate measurement of characteristic compounds formed in the body. 1-Hydroxypyrene is widely used as a urinary biomarker of exposure to polycyclic aromatic hydrocarbons. Common analytical approaches, however, often require chromatographic instrumentation and sample pretreatment, creating a need for simpler methods suitable for measurements closer to the point of use.
Cyclodextrins are ring-shaped sugar molecules with central cavities that can accommodate selected molecular guests. This study examined structurally different cyclodextrin platforms as molecular-recognition scaffolds for sensitive and selective fluorescence detection of 1-hydroxypyrene.
Comparing Cyclodextrin Architectures
Veclani and colleagues evaluated six cyclodextrin-based hosts, including monomers, polymers, and dimers. The study considered not only cavity size but also how multiple cyclodextrin rings can cooperate in guest binding.
- Aim: To identify cyclodextrin architectures that bind 1-hydroxypyrene strongly and selectively while producing fluorescence behavior suitable for detection.
- Materials: Hydroxypropyl-beta-cyclodextrin, gamma-cyclodextrin, beta- and gamma-cyclodextrin polymers, and two structurally different beta-cyclodextrin dimers were studied.
- Experimental approach: UV–visible absorption, steady-state fluorescence, and time-resolved fluorescence were used to determine binding stoichiometry, affinity, and emissive states.
- Computational approach: Molecular-dynamics trajectories of 100 nanoseconds and thermodynamic-integration binding-free-energy calculations were compared with the experimental affinity hierarchy.
Fluorescence Changes with Molecular Assembly
The binding ratio and fluorescence response varied according to cyclodextrin architecture.
- Different assemblies: Host and guest formed not only 1:1 complexes but also 2:1 and 2:2 assemblies. Binding constants for the 1:1 complexes ranged from 10³ to 10⁵ M⁻¹.
- Strong 1:1 binding: Hydroxypropyl-beta-cyclodextrin showed the highest 1:1 affinity, with a binding constant of 1.0 × 10⁵ M⁻¹.
- Gamma-cyclodextrin behavior: Gamma-cyclodextrin formed cooperative 2:2 assemblies with pyrene-excimer emission at 480 nm, indicating close interaction between excited molecular guests.
- Recognition by a dimer: One beta-cyclodextrin dimer promoted deprotonation of 1-hydroxypyrene upon complexation, producing a fluorescence spectrum distinct from the free compound and from other common metabolites.
- Computational agreement: The simulations and free-energy calculations reasonably reproduced the experimentally observed order of host affinities.
Selective Detection in the Nanomolar Range
Preliminary testing with the selected beta-cyclodextrin dimer indicated the potential for selective measurement of 1-hydroxypyrene at low concentrations.
- Measurement conditions: 1-Hydroxypyrene was excited at 400 nm in the presence of 1 mM cyclodextrin dimer.
- Response range: A linear fluorescence response was obtained from 10 to 1000 nM.
- Selectivity: Detection was not disrupted by the competing metabolites 1-naphthol, 2-naphthol, 2-fluorenol, or 9-fluorenol.
- Importance: Arranging cyclodextrin rings as dimers or polymers makes it possible to design both molecular binding and the resulting fluorescent state.
- Potential application: The approach may support fluorescence-based monitoring of environmental-exposure biomarkers with less dependence on extensive sample preparation and sophisticated instrumentation.
Overall Significance
The study shows that cyclodextrin sensing performance is determined by more than cavity size. Monomeric, polymeric, and dimeric architectures alter guest orientation, assembly stoichiometry, intermolecular distance, and acid–base state, which in turn change fluorescence intensity and spectral position.
The 2:2 assembly formed by gamma-cyclodextrin illustrates how cooperative molecular recognition can create a new emissive state. The beta-cyclodextrin dimer, meanwhile, produced a promising response for distinguishing 1-hydroxypyrene from related metabolites. Controlling the spatial arrangement of cyclodextrin cavities may therefore provide a useful route to sensitive and selective molecular sensors.
