HPβCD for Smart Delivery and Water Treatment: Molecular Technologies for Solubilization, Transport, and Separation
Introduction
2-Hydroxypropyl-β-cyclodextrin has a water-compatible exterior and a relatively hydrophobic molecular cavity. This structure can encapsulate poorly soluble drugs and regulate molecular movement within functional materials. The studies highlighted here use HPβCD to improve an antibacterial candidate, deliver two drugs through the skin to scar tissue, and engineer a drinking-water membrane that removes trace pollutants while allowing mineral ions to pass.
Across these applications, the HPβCD cavity acts not only as a molecular container but also as a nanoscale environment that controls molecular state and transport. It supports solubilization and stabilization in drug systems, responsive release in hydrogels, and selective diffusion inside membrane pores.
Research Highlights
1. Improving the Solubility and Particle Uniformity of a Fluoroquinolone Candidate
- A poorly soluble novel fluoroquinolone molecule, PFQA, was studied in the presence of HPβCD.
- HPβCD increased PFQA solubility by up to approximately twofold.
- Drug particles were reduced to sizes of up to about 100 nm, with substantially improved population homogeneity.
- Infrared spectroscopy and proton nuclear magnetic resonance were used to investigate complex formation.
- PFQA exhibited two binding sites for HPβCD.
- The results suggested possible negative cooperativity, in which occupation of one site sterically reduces binding at the other.
2. pH-Responsive Transdermal Co-Delivery to Hypertrophic Scar Tissue
- Triamcinolone acetonide and 5-fluorouracil were encapsulated with HPβCD to improve aqueous solubility and stability.
- The inclusion complexes were incorporated into a borax-cross-linked dextran hydrogel.
- The system released drugs rapidly in the weakly acidic hypertrophic-scar microenvironment and enhanced transdermal absorption and cellular uptake.
- It promoted a shift from pro-inflammatory M1 macrophages toward the repair-associated M2 phenotype.
- Abnormal activation of scar fibroblasts and expression of fibrosis-related proteins were reduced.
- Cell and animal studies demonstrated anti-inflammatory and antifibrotic effects.
3. A High-Permeance Membrane That Retains Pollutants but Passes Minerals
- A polyamide nanofiltration membrane was modified through HPβCD doping and secondary sulfonamidation with sulfuryl chloride.
- The membrane reached a water permeance of 26.9 L m⁻² h⁻¹ bar⁻¹, 3.5 times that of NF90 and 2.4 times that of NF270.
- It strongly rejected five representative emerging pollutants while selectively permitting passage of dietary mineral ions.
- A thinner, more hydrophilic active layer promoted water transport, while a more negative surface charge favored mineral-ion passage.
- Tailored pore size and HPβCD-enhanced intrapore diffusion resistance supported broad-spectrum pollutant rejection.
- Potential uses include secondary water-supply treatment and point-of-use purification.
Overall Significance
These studies show that HPβCD functions beyond its conventional role as a pharmaceutical excipient. It serves as a molecular transport-control component that can improve formulation of poorly soluble compounds, support site-responsive local therapy, and balance permeability with selectivity in water-treatment membranes.
Potential applications include formulations for difficult-to-dissolve drug candidates, minimally invasive skin treatments, and purification systems that retain useful minerals. Further development requires application-specific validation of efficacy and safety, long-term storage stability, membrane fouling resistance during continuous operation, and reproducible quality in large-scale manufacturing.
