β-Cyclodextrin Polymers for Drug Delivery, Water Safety, and Environmental Remediation
Introduction
β-Cyclodextrin is a cyclic oligosaccharide with a molecular-scale cavity. When this molecular-recognition function is integrated into fibers, cross-linked polymers, or composite materials, it can support sustained drug release and the removal of contaminants or pathogens from water.
The studies highlighted here address a fibrous insert for sustained ocular delivery, a quaternized cyclodextrin polymer targeting bacterial cell envelopes, and recent developments in β-cyclodextrin composites for wastewater treatment. They share a common strategy: combining host-guest recognition with the form, stability, and processability of polymer materials.
Research Highlights
1. Polymer-Free Fibrous Inserts for Sustained Ocular Delivery
- Poorly water-soluble fenofibrate was complexed with hydroxypropyl-β-cyclodextrin.
- Uniform fibrous inserts were produced by electrospinning without adding a separate polymer.
- Fiber diameters ranged from 2.04 to 2.75 µm, and drug loading reached 8.56% by weight.
- Increasing drug loading reduced the complexed fraction from 74% at 2.5% loading to 23% at 10% loading.
- Drug release continued for 168 hours with a burst release below 2%.
- Ex vivo transscleral testing showed 0.35–0.5% penetration within 24 hours, compared with undetectable penetration for free fenofibrate.
- Cell viability remained above 85% for all formulations, and irritation testing showed minimal effects.
- Cell-based angiogenesis studies demonstrated dose-dependent biological activity.
2. A Cyclodextrin Polymer Targeting Bacterial Cell Envelopes
- Quaternary-ammonium-functionalized cyclodextrin units were cross-linked into a β-cyclodextrin oligomer network.
- Positively charged and natural cyclodextrin sites were coordinated within one material.
- Binding to five major bacterial cell-envelope components was evaluated.
- Effects on inactivation kinetics followed the order lipopolysaccharide, peptidoglycan, cardiolipin, and phosphatidylglycerol approximately equal to phosphatidylethanolamine.
- The material achieved 1.7–9.2 log inactivation against E. coli and several drug-resistant ESKAPE pathogens.
- Performance was demonstrated in both simulated and natural water.
- A packed column maintained more than 96.8% E. coli removal after treatment of 200 pore volumes of natural water.
- The study provides a design strategy for broad-spectrum, long-lasting antimicrobial water-treatment materials.
3. Mapping β-Cyclodextrin Composites for Wastewater Treatment
- The review assessed β-cyclodextrin water-treatment materials reported from 2021 to 2026.
- Target contaminants included pharmaceuticals, dyes, pesticides, heavy metals, PFAS, and other emerging pollutants.
- Immobilization and cross-linking were compared as solutions to the limited recovery, stability, and reuse of free β-cyclodextrin.
- Composite partners included polymers, biomass, carbon materials, magnetic particles, MOFs, COFs, photocatalysts, and membranes.
- Removal mechanisms included inclusion, interfacial interactions, adsorption, photocatalytic degradation, and membrane separation.
- Density-functional theory, molecular dynamics, molecular docking, and artificial intelligence were discussed for interpretation and prediction.
- Practical challenges include standardized testing, continuous-flow and real-wastewater validation, leaching, regeneration waste, cost, scale-up, and life-cycle assessment.
Overall Significance
Together, these studies show that β-cyclodextrin polymers are evolving from molecular containers into functional platforms that control release time, bacterial-interface interactions, and selective pollutant removal. Ocular inserts combine inclusion with fiber formation. Antimicrobial materials combine positive charge with a polymer network. Water-treatment composites add recoverability, durability, and separation functions to molecular recognition.
Future development should evaluate long-term ocular safety and efficacy, continuous operation in natural and real wastewater, material regeneration and reuse, component leaching, and scalable manufacturing. β-Cyclodextrin polymers offer a shared material concept for retaining useful molecules and selectively removing harmful targets in medical and environmental applications.
